CUTICULAR PROTEINS AND THEIR AMINO ACID COMPOSITION IN LAST LARVAL CYCLORRHAPHA (DIPTERA)

Abstract:
Two-dimensional isoelectric focusing SDS-PAGE of urea-extractable cuticular proteins of the last larval Cephalopina titillator was done and compared with other larval cyclorrhaphans. The proteins usually have isoelectric points ranging from 4.5 to 8 in the early (EL) and from 4.5 to 9 in the late (LL) last instar. In LL new sets of proteins are formed, usually of higher isoelectric points. The amino acids composition of the extracted proteins of C. titillator , Gastrophilus intestinalis , and Parasarcophaga aegyptiaca were estimated. The profiles of the amino acid composition in both the EL and LL of these cyclorrhaphans usually have the same trend; therefore, were considered and discussed as one pool. In these cyclorrhaphous larvae the predominant residues are proline and glycine that represent ~50 %. The acidic residues glutamic and aspartic represent ~18 %, and the basic residues histidine, lysine and arginine represent ~9 %. The hydrophobicity is always higher in the LL than in EL. These characteristics are discussed.
