RECOGNITION OF ECDYSONE BINDING SITES IN THE FAT BODY CELL MEMBRANES OF THE SILKWORM BOMBYX MORI

Abstract:
In the present study, we detected the membrane ecdysone-binding sites in the plasma membrane of fat body cells of the silk worm Bombyx mori . Membrane fractions prepared from fat body, during different days after gut purge, exhibit different binding activities to ponasterone A (25-deoxy-20-hydroxyecdysone) (PonA). Membrane fractions from day 0 (G0) larvae showed the highest binding activity, and were accordingly used for saturation analysis. G0 membrane fractions did not contain conventional nuclear ecdysone receptor as revealed by Western blot analysis using antibody raised against B. mori nuclear ecdysone receptor B1 (BmEcR-B1). The saturation analysis showed the presence of a single high affinity binding site of high affinity to PonA with an apparent Kd of 15.1× 10-9 M; which is in good accordance with the Kd value we previously obtained for membrane ecdysone receptor (mEcR) from anterior silk glands (ASGs). The maximum receptor number (Bmax) in fat body membranes was shown to be 0.42 pmol.mg-1 protein.
