EXOPEPTIDASES ACTIVITY IN THE MIDGUT REGIONS OF THE LARVAL PARASARCOPHAGA HERTIPES (SARCOPHAGIDAE, CYCLORRHAPHA, DIPTERA)

Abstract:
Extracts of the anterior- and posterior midgut regions of larval Parasarcophaga hertipes exhibited leucine aminopeptidase-like (LpNA hydrolyzing), carboxypeptidase A-like (HpLA hydrolyzing), and carboxypeptidase B-like (HA hydrolyzing) activities. These activities showed maxima at pHs between 8.5-9.0, 7.0-9.0, and at pH 7.0 for these enzymes, respectively. The unenhanced leucine aminopeptidase-like activity of the two regions was inhibited by PMSF, and that of the posterior midgut only, was enhanced by Zn2+, Ca2+, and Mn2+. The enhanced and the non-enhanced (only after preincubation with divalent cations) activities were inhibited by the cation chelator 1,10-phen, but not by EDTA. Carboxypeptidase A- and B-like enzymes require Zn2+, Ca2+, or Mn2+, to display their activity, and were inhibited by 1,10-phen, but not by EDTA. The presence, distribution in the midgut regions, and characterization of these exopeptidases are discussed.
