Abstract:
The extractable cuticular proteins (using urea/SDS, at pH 8.6) of Parasarcophaga aegyptiaca (Salem) were characterized during development from the third-instar larvae up to the adult. One-dimensional electrophoresis of SDS-PAGE for the extractable cuticular proteins shows that major protein bands of lower and medium molecular weights in the larval and pupal stage (puparia) are common and nearly identical. The essential nature of these proteins in cuticle structure is proposed. Other proteins are found predominantly in a particular instar or at a particular period within an instar. Adult proteins reveal a different set of major and minor proteins. Two-dimensional electrophoresis of the extractable cuticle proteins shows that the epidermal cells start expression of a new set of cuticular proteins in the late pupal stage (puparia). It is revealed also that most cuticular proteins of P. aegyptiaca larvae are mainly acidic like other flexible, soft-bodied larvae of Diptera and Lepidoptera. Major proteins extracted from pupae are slightly acidic, and that of the early pupae are neutral to slightly basic. Cuticular proteins of adult and, somewhat, the late pupae are mainly basic resembling cuticular proteins of the hard-bodied insects.
