Abstract:
The chitin-synthesis inhibitor lufenuron was administered to Drosophila melanogaster by incorporation of the LC50 (12 ppm) into the larval rearing medium to reveal its effects on the induction of sex-linked recessive lethals (SLRL), asynapsis in polytene chromosomes, and chromosomal aberrations in mitotic cells. The results show that lufenuron increases the frequency of SLRL, as evaluated by the use of Muller-5 (M-5) marker, in the three successive broods that represent the three successive stages of spermatogenesis, i.e. spermatozoa, spermatids, and spermatocytes. In the salivary gland cells, lufenouron induced asynapsis of the polytene chromosomes to a significantly higher level in both parents (P) and the second generation (F2) than that of the spontaneous one. In neuroblasts of the brain, lufenuron induced chromosomal aberrations of the mitotic chromosomes of both P and F2. These findings suggest that leufenuron is a potent mutagen to both germ-line cells and somatic cells of D. melanogester .
